Why the cell source matters more than the brand

Cell type, donor, tissue of origin and passage number change what a preparation contains. None of these are visible on a box.

The short answer

What a cell releases depends on what kind of cell it is, which tissue and donor it came from, how many times it has been divided in culture, and the conditions it was grown under. Every one of those variables changes the composition of the resulting preparation.

None of them appear on most product packaging, and several are treated as commercially confidential. A brand name identifies a supplier. It does not identify a cell, and two products from the same supplier can differ if the cell bank or the culture protocol changed.

The variable nobody prints on the box

Ask what determines the contents of a cell-derived preparation and the answer runs, in rough order of influence: the isolation method, the culture conditions, the cell type, the donor, and the passage number. Cell type

Different cell types have different secretomes. That is not a subtle effect. A fibroblast, an adipose-derived stromal cell, an umbilical cord-derived cell and a keratinocyte release substantially different sets of proteins and vesicles, because they are different cells doing different jobs. A preparation from one is not interchangeable with a preparation from another, and there is no general principle establishing which source is preferable for skin.

Tissue and donor

Within a cell type, the tissue it was taken from and the individual it came from both matter. Donor variation in cell behaviour is a well recognised practical problem in cell culture. A supplier using a single donor cell bank has consistency within that bank and a discontinuity when the bank is replaced. A supplier pooling donors has averaged variation and less traceability.

Passage number

Cells change as they are repeatedly divided in culture. Growth rate, morphology and secretory profile all shift over passages, and at high passage numbers cells may enter senescence, at which point their secretory profile changes markedly. Manufacturers therefore work within defined passage windows. A product specification that does not state the passage window has omitted a variable known to change the product.

Culture conditions

Oxygen tension, medium formulation, whether serum was used and whether it was vesicle depleted, cell density at harvest, and time between medium change and collection all influence what ends up in the conditioned medium. Some of these are chosen to maximise yield, and yield-maximising conditions are not necessarily composition-preserving conditions.

Human, animal and plant sources

Preparations in this market are described as derived from human cells, from animal cells, or from plant material. These are not variations on one theme. They carry different biological plausibility, different safety questions and different regulatory consequences.

  • Human-derived. Most plausible for signalling into human tissue, since receptor and ligand systems match. Raises human tissue questions covered in human tissue considerations, including consent, donor screening and traceability.
  • Animal-derived. Raises questions of cross-species signalling compatibility and of viral safety, and generally requires the manufacturer to explain why signalling molecules from another species should engage human receptors usefully.
  • Plant-derived. Plant cells do release vesicle-like particles, and this is a genuine research area. Plants do not use the growth factor and cytokine systems that animal cells use, so a plant-derived preparation cannot be argued to work by delivering those molecules. Any mechanism proposed must be a different one, and should be stated.

The point is not that any of these is invalid. It is that they need different arguments, and the same marketing language is applied across all three as though the argument were the same.

Immortalised lines and the consistency trade

Some manufacturers use immortalised cell lines rather than primary cells, because a line can be grown indefinitely and provides consistency that primary cells cannot. The trade is that an immortalised line has been altered to divide without limit, which is a change to the cell's growth control, and a preparation derived from such a line should be described as such.

This is a legitimate manufacturing choice with a real advantage. It should also be disclosed, because it bears on both the biological argument and the regulatory analysis. A supplier that discloses it is telling you they have thought about consistency. A supplier that does not disclose the cell system at all has not given you enough to think with.

Why brand is a weak proxy

A brand tells you who sells the product. It does not tell you who manufactured it, whether the manufacturer changed, whether the cell bank was replaced, or whether the process was modified. In markets where products are white labelled, several brands may share a manufacturer and differ only in packaging and price.

That is not necessarily improper. It does mean that comparing brands on the basis of brand material compares marketing. The comparison that would matter is between manufacturing specifications, and those are the documents least often shared. Our listings page exists partly for this reason: it accepts only verifiable, disclosed facts about sourcing and provenance, and it publishes what a supplier declines to disclose alongside what they do.

What a good disclosure looks like

We would consider a supplier well disclosed if a purchaser could learn, without signing anything: the species and cell type, the tissue of origin, whether the bank is single donor, pooled or immortalised, the passage window, whether serum is used and whether it is depleted, the isolation method by name, and the characterisation performed on each batch.

That is eight facts. None of them is a trade secret in any meaningful sense: they are the description of a process, not the process itself. A sector in which eight facts are considered confidential is a sector where the purchaser cannot evaluate what they are buying, and that condition, rather than any particular product, is the thing worth objecting to.

Does the cell source change the product?

Yes. Different cell types release different sets of proteins and vesicles, and donor, tissue of origin, passage number and culture conditions all add further variation. Source is one of the main determinants of what is in a preparation.

Are human-derived products better than animal or plant derived?

Human-derived material has the most straightforward biological argument for signalling into human tissue, because receptor and ligand systems match. Animal and plant sources require a different mechanistic argument, which should be stated rather than assumed.

What is passage number and why does it matter?

It is the number of times a culture has been subcultured. Cells change with passage, including what they secrete, and at high passage they may become senescent with a markedly different profile. A specification without a passage window has omitted a known variable.

Why does serum in the culture medium matter?

Serum contains its own vesicles, lipoproteins and large amounts of protein that overlap with the material being isolated. Unless serum-free or vesicle-depleted conditions are used, some of the final product came from the serum.

Can I compare two products by brand?

Not usefully. Brands may share manufacturers, may change manufacturers, and rarely publish the process facts that would allow comparison. A comparison of manufacturing specifications would be informative; a comparison of brand material is a comparison of marketing.
